Training
You MUST have attended the Introduction to Flow Cytometry lecture series before signing up for Training - New Users, CSC.
Once you have attended the Introduction to Flow Cytometry, make an appointment for:
01. Training - New Users, CSC
Tuesdays, 9-11am, CSC K4/535
This 2-hour, hands-on training on our benchtop cytometers is intended for those with little or no flow cytometry experience. This class is designed to teach the basics of data collection and management. No personal samples are used.
Analysis Computers
Off-line data analysis computers are available to facility users only and are located at both the CSC location and at the McArdle laboratory. Computers are loaded with a variety of analysis packages including FlowJo, CellQuestPro, FCS Express, ModFit, WinList, Image analysis, as well as standard Office software.
There is no charge for the use of these computers but are exclusively for analysis of data generated at this facility.
You may reserve a computer on-line, but they are also available on a first-come, first-served basis.

Benchtop Analyzers
The benchtop instruments used for this purpose are the FACSCalibur
(488nM and 633nM lasers) or the LSRII (488nm, 405nm, UV, and
633nm lasers).
When signing up for appointments here, insure all fluorochromes
to be used can be excited with the lasers listed. The instruments
are available for self-run appointments for which no assistance
is provided, or for staff-assisted appointments for those who
need assistance in data collection or instrument operation. Staff-assisted
appointments can be used by those who have not received any instrument
training but need samples analyzed, as well as by those who need
assay-specific instruction.

Sorters
The facility has two high-speed sorters: one located in the McArdle facility and one at the CSC location. Each sorter has some unique capabilities for the maximum applications, but they both can sort up to 4 separate populations simultaneously. Operated by staff ONLY. They are both housed under BSL2 conditions. Note: Samples exceeding BSL2 levels when aerosolized cannot be sorted!!

This FACSVantageSE is equipped with 3 lasers (488 nM solid state blue, 405 nM solid state violet, and 633 nM solid state red) and digital electronics (DiVa). Like the LSRII it has variable emission filter sets (11 options). Please check with us if you have questions about fluorochrome selections for this sorter.
12. Sorter-McArdle
This FACSVantageSE with 3 lasers (argon, tunable—488, 514, 457, etc.; krypton tunable—UV, 405, 413, 568 etc.; 633 nM HeNe) and digital electronics (DiVa). Because this sorter has flexibility in excitation and emission wavelengths, please check with us if you have questions about fluorochrome selections for this sorter.
Microscopes
The Pathway is a microscope with a temperature-controlled chamber and liquid handling capabilities. This inverted fluorescent scope requires slides or 96/384 well microtiter dishes. It is equipped with two mercury arc lamps, 16 different excitation filter sets and 8 different emission filter sets. It uses standard immunofluorescent probes, like Texas Red, Hoechst, and the Alexa dyes and can collect either regular or confocal images.
